This study systematically explored the regulatory mechanism of soil environmental factors (pH, humidity, temperature, and organic matter content) on the colonization ability of Penicillium purpurogenum and its nematode control effect. The experiments showed that within the pH range of 6.0-7.5, the spore ger-mination rate of Penicillium purpurogenum reached over 92%. The optimal humidity for mycelial expansion was 25%-30% of field capacity, with a growth rate of 4.3mm/d. When the temperature was maintained at 25-28℃, the nematode parasitism rate increased by 47.6% compared to the control group. When the soil organic matter content was ≥2.5%, the spore production of Penicillium purpurogenum increased by 2.1 times, and it could continuously release chitinase (activity up to 38.7U/mg) to destroy nematode egg shells. Principal component analysis revealed that the interaction of the four factors could explain 78.3% of the variation in control effect (R²=0.783, p<0.01). This study provides a theoretical basis for constructing a three-in-one biological control model of "soil micro-environment-microbial activity-pest control", which is particularly suitable for green control of root-knot nematodes in protected agriculture. It is expected to reduce the use of chemical pesticides by more than 60%, and has important practical value for achieving ecological agricultural construction under the "dual carbon" goals.
